Functional suppression of neutrophils against candida albicans during Friend leukemia virus infection in mice and restoration by interleukin 8 (IL8) and bacterial lipopolysaccharide (LPS).

Retroviral infection of mice with Friend leukemia virus (FLV) is widely known to cause suppression of function associated with T cells, B cells, NK cells, and macrophages. We now show that such retroviral infection can also functionally suppress murine neutrophils against C. albicans, which is an opportunistic fungal pathogen commonly encountered in AIDS patients. Balb/c mice were inoculated with proteose peptone 4 hr prior to harvesting of the elicited neutrophils from the peritoneal cavity. The neutrophils, incubated with C. albicans, for 18 hr at 37C, were highly effective against Candidal growth as measured by a 3H-glucose incorporation assay. Infection with FLV, however, caused a time-dependent loss of antifungal activity in the elicited neutrophils. This defect was detectable at 3 days after infection and became maximal by day 14. To define if the suppression of function was permanent or reversible, the effect of Escherichia coli LPS, a strong neutrophil activator, and IL8, a recently identified chemotactic and activating cytokine for human neutrophils, were tested. Similar to man, treatment of normal murine neutrophils with LPS or IL8 was found to significantly enhance antifungal activity above baseline controls. More importantly, these two immunomodulators could restore antifungal function in neutrophils from FLV-infected mice. Functional restoration was obtained even with neutrophils from mice that had been totally immunosuppressed after 28 days of infection. These results indicate that FLV is a potent neutrophil suppressing virus but that this negative property of FLV can be overcome by IL8 or LPS.
