Sample type RNA

Channel 1
Source Name wildtype mice
Organism(s) Mus musculus
Characteristics strain: C57Bl/6
gender: male
age: 10 months
tissue: whole heart
Biomaterial provider Cecilia Hertig, INGEBI-CONICET, Vuelta de Obligado 2490, 1428 Buenos Aires
Treatment protocol No treatment was applied.
Growth protocol Animals were kept in a SPF barrier under standardisized conditions with respect to temperature and humidity, and were housed on a 12h light/12h dark cycle in groups of 4-5 with free access to food and water. Animal housing, care, and applications of experimental procedures complied with the Guide for the Care and Use of Laboratory Animals of the Government of Berlin, Germany and are in accordance to the recommendations of the Society for Laboratory Animal Science (GV-SOLAS) and the Federation of European Laboratory Animal Science Associations (FELASA).
Extracted molecule total RNA
Extraction protocol Total RNA was isolated using TRIzol Reagent (Invitrogen) according to manufacturer`s instructions.
Label Cy3
Label protocol Labeling of the targets during reverse transcription was performed as described by Pat Brown (http://brownlab.stanford.edu/).

Channel 2
Source Name wildtype mice treated with Doxorubicin
Organism(s) Mus musculus
Characteristics strain: C57Bl/6
gender: male
age: 10 months
tissue: whole heart
Biomaterial provider Cecilia Hertig, INGEBI-CONICET, Vuelta de Obligado 2490, 1428 Buenos Aires
Treatment protocol Wildtype mice were treated 3 times a week with 15mg/kg Doxorubicin.
Label Cy5
Label protocol Labeling of the targets during reverse transcription was performed as described by Pat Brown (http://brownlab.stanford.edu/).

Hybridization protocol Targets were vacuum-centrifuged and resuspended in 50 μl DIG hybridisation buffer (Roche). Denatured targets (98°C, 2 min) were hybridised in a humidified chamber at 42°C overnight. The following day arrays were washed (2xSSC; 0,03% SDS for 10min, 2xSSC for 5min, 1xSSC for 5min, 0,2 x SSC for 2min) and dried in a centrifuge.
Scan protocol Agilent Microarray scanner (G2565BA)
Description The gene expression of the whole heart of Doxorubicin treated wildtype mice was compared with wildtype mice.
Data processing Local background was subtracted and LOWESS normalisation was performed using Feature Extraction Software (version A 6.1.1).
