Anti-Protective Antigen (PA) of B. anthracis IgG Subclasses Distribution in Anthrax Vaccine Adsorbed (AVA) Recipients.

BACKGROUND: Correlates of human protection against anthrax have yet to be fully elucidated. Protective Antigen (PA) is the pivotal protein of the anthrax toxin complex and the principal immunogen of licensed and research anthrax vaccines. Anti-PA immunoglobulin (IgG) antibody levels are considered an appropriate marker of human immune responses to anthrax vaccines and Bacillus anthracis infection. Studies on guinea pigs have indicated that anti-PA IgG subclass distribution may correlate with survival against challenge. In humans, however, there is limited information about IgG subclass distributions in the serum of anthrax vaccine recipients or persons infected with B. anthracis. METHODS: A quantitative Enzyme Linked Immunosorbent Assay (ELISA) for evaluation PA specific IgG[1], IgG[2], IgG[3] and IgG[4] subclass distributions in humans vaccinated with AVA has been developed. Anti-PA IgG subclass concentrations were assigned for anti-AVA Human Reference Serum AVR414 and used as a standard to quantify anti-PA IgG subclasses concentrations in serum from 27 healthy adult volunteers who had received different numbers of doses of AVA. RESULTS: Analysis of subclass distributions showed that Protective Antigen induces all four IgG subclasses with a predominant IgG[1] response (mean 54.18% of total IgG, 95% CI: 47.96-60.4). The percentage of IgG[2] and IgG[4] was similar (mean 23.17%, 95% CI: 21.14-25.22 for IgG[2] and mean 19.93%, 95% CI: 12.06-27.81 for IgG[4], respectively). The concentration of IgG[3] was low (mean 3.7% of total IgG, 95% CI: 2.83-4.79). CONCLUSIONS: These data indicate that anti-PA IgG subclass distribution in response to AVA immunization is IgG[1], IgG[2] IgG[3]and IgG[4] subclasses.
