Which cell type in semen is the major source of HIV-DNA?

HIV may be spread by sexual intercourse from male to the partner by free virus in the ejaculate plasma, by HIV absorbed to sperm or by HIV harboured in the contaminating round cells. OBJECTIVE: To find out whether round cells or spermatocytes are the major sources of HIV. Ejaculate was separated into plasma, round cells and spermatocytes by ficoll centrifugation. Round cells are characterized by flow cytometry. DNA was liberated by proteinase-K digestion and amplified by PCR using the SK68/69 primers. Blood lymphocyte (PBL) DNA was used as controll. RESULTS: In all of 39 HIV infected subjects HIV-DNA was detected in PBL, in 8 (20.5%) HIV-DNA was identified in the round cell-DNA and in no sample of the spermatocyte-DNA. Fractions from 10 HIV uninfected individuals were always negative. CONCLUSION: The major source of HIV-DNA in the ejaculate is the round cell fraction, while spermatocytes seem to be free of HIV-DNA.
