Comparison of two HIV-1 genotypic analysis softwares ; HGS version 2.0 and HRP-ASAP version 10.

Background: HIV genotyping is an indirect antiretroviral drug resistance testing. It is difficult to translate the results of a genotypic resistance test into a meaningful conclusion about the resistance of the virus to drugs. Thus the genetic mutation databases are important in the genotypic resistance interpretation. This study aim to compare the genotypic mutation databases of HGS version 2.0 and HRP-ASAP version 1.0. Methods: Population sequencing of the entire protease and first 320 codons of reverse transcriptase from HIV-1 infected Thai patients were performed using the HIV Genotyping System Kit, followed by analyzing the amino acid substitution that difference from the wild type by HGS version 2.0 and HRP-ASAP version 1.0. Results: Thirteen mutations conferring drug resistance protease gene at position L10Y(2%), K20T(2%), L33V(4%), L33I(2%), F53L(6%), I54A(2%), L63C(2%), L63A(2%), T74S(6%), T74A(2%), I85V(2%) and I93L(10%), and 8 mutations conferring drug resistance in reverse transcriptase gene at position K43E(78%), E44D(4%), V75M(4%), A98S(10%), V118I(4%), Q151M(6%), H208Y(4%) and L228H(8%) were detected and identified by HRP-ASAP version 1.0. Though, HGS version 2.0 could detect all of these 21 codons, however the program has identified these mutations as novel nucleotide variance. Conclusion: HGS software version 2.0 may contain lists of the resistance mutations known at the time of the software's release while HRP-ASAP 1.0 has been updated regularly through the internet. Therefore, HRP-ASAP 1.0 is more reliable and suitable program for detection of resistance mutations in HIV-1 polymerase gene.
