Use of the Yellow Fever Virus Vaccine Strain 17D as a Model to Investigate Hepatitis C Virus Inactivation Efficacy in Gastrointestinal Endoscopy.

BACKGROUND: Transmission of HCV through endoscopy has been reported, mostly related to an inappropriate disinfection procedure. As HCV is a non-cultivable virus, experimental investigation of the ability to eradicate HCV from contaminated endoscopes are missing. We describe a new approach using the cultivable attenuated yellow fever virus vaccine strain 17D (YF17D) as a model of HCV contamination. Material and method. YF 17D strain and a live attenuated vaccine strain of poliovirus, boosted to high titer by passage through vero cell culture were used. 15 ml of a suspension was flushed through the four channels of a gastroscope or a colonoscope. A thorough manual cleaning-washing disinfection procedure was then performed using alkaline detergent alone or followed by a 2% glutaraldehyde solution. Residual viruses were harvested by an exhaustive flush of a recovery solution. Quantitative virus assays were performed in a 50% tissue culture infective dose assay after concentration of the samples by tangential ultrafiltration.RESULTS: (Mean log of TCID 50%). Conclusion: This study demonstrates that live attenuated yellow fever virus vaccine strain could be easily used as an alternative model for the evaluation of the strategies designed to reduce the transmission of HCV by complex medical device. The low residual burden observed after disinfection contrasts with the negative data obtained after PCR detection of HCV in other studies.KEYWORDS: Disinfectants; Endoscopy; HCV
