Susceptibility of the human foamy virus (HFV) to clinically used antiretroviral drugs.

The Human Foamy Virus (HFV) is a member of the Retroviridae family belonging to the spumaretrovirus genus. Although HFV is less frequently implicated than HIV and HTLV in human diseases, some recent studies had suggested its association with the Basedow-Graves Disease and some other autoimmune diseases such as myasthenia gravis. Gill et al (N.E.J.M. 1995) showed the interest of a treatment by zidovudine associated with interferon alpha (IFN alpha) in the evolution of Adult T-cell leukemia-lymphoma. In the same way, a treatment by an antiviral drug alone or in association could be useful in the treatment of diseases suspected to be associated with the HFV. Our aim was to determine the susceptibility of the HFV to several antiviral drugs, known for their activity against the replication of HIV. The results, in term of Inhibiting concentrations IC(50) and IC(90), are summarized in the following table:(Tabular Data: See Abstract Volume). In conclusion, we demonstrated that almost ZDV, ddC and d4T are effective against HFV replication. However, ddI and TIBO R82913 don't seem to be effective in a range of concentrations compatible with a clinical use. We are now determining the IC of some antiprotease drugs and of Lamivudine (3TC). After further investigations, the differences of the activity of the reverse transcriptase (RT) inhibitors against the HFV and the HIV replications would be correlated with the differences in the respective protein sequences of the RTs.
