  We have set up the RNase I11 assays in crude g. coli extracts
(S 30) essentially as described by Robertson, Webster, and Zinder,
J.B.C. w:
We have the C labeled. We have done this both with some extracts and
with alumina ground cells. The assay has 0.1 NfIccC1, 0.01 (Mg(Ad2,
0.02 g Tris pH 7.L, 50 mules of polper P per nl.  We use a 0.5 nzl
assay-mix because our poly C is not very hot.  We 5% TCA and add BSA]
carrier.
I don't: like locking for disappearance so we fall= formation oE acid
soluble cptn.

83 (1968), except that we use poly I'poly C as substrate.

They counted filters after filtration on W/A fiber filters.

is to slice gals, ~?laslt each slice in a litth buffer, sic in cold for
a w1ii.h (Zi Izx to overnight) centrifuge off gal and do assay.  Gels can
be nicely sliced info 1-2 rn pieces by band with a razor or sscalpal iE
tkay are frozen and kept so with yawd@rad dry ice during siicing.

So experience with iV4aEze assays on gels.
