Multiple herbal therapy: a new approach into antiviral treatment. 1. In vitro activity.

OBJECTIVES: To develop a new herbal extract with antiviral and immunological properties. This objective was achieved using a new approach by combining three herbs. The idea behind this approach was to increase the potency of the extract and to minimize some of the already known side effects exhibited by some of these herbs. RESULTS: A novel extract was obtained from mixing N. oleander, V. album and A. graveolens. Some of the major components of the extract was identified and characterized using some qualitative and quantitative analysis like TLC, HPLC, 1HNMR, 13CNMR ... etc. When the extract was tested against the reverse transcriptase enzyme in biochemical assay, it showed an ID50 of about 9-10 ug/ml. Further, when assessed against HIV-I in vitro, using a standard, syncytial reduction assay. The results showed a significant reduction to syncytia [45%] compared to virus control at conc. 0.2-2 ug/ml. The extract also showed significant RIP activity in in vitro system at conc. less than 1 ug/ml. On the other hand, the extract demonstrated astrong inhibition of the complimentary activity (CP) CPIC50 was found to be 5.2 ug/ml. Other tests on the primary humoral immune response in mouse spleen cells cultured with sheep erythrocytes indicate an immune-modulatory behaviour of the extract. CONCLUSION: The above results indicate significant antiviral activity in vitro in addition to the immuno-modulatory characters of the extract, such results are encouraging to carry further preclinical studies.
