Photoinactivation of HIV-1 by dimethylmethylene blue.

Background: Phenothiazine dyes can photoinactivate viruses due to their property of releasing singlet oxygen and affinity for nucleic acid and proteins when activated with light. Dimethylmethylene blue (DMMB) yields approximately 50% more singlet oxygen than methylene blue (MB) and has a higher affinity for nucleic acids. DMMB has been shown to inactivate various model viruses with minimal alternations of RBC properties.Methods: Analogous to the model viruses, we used DMMB and light to photo-inactivate intra- and extra-cellular HIV in blood products. Plasma, Packed Red cells and whole blood were experimentally inoculated with 2000 TCID50 of primary HIV-1 isolate (UG/92/021), incubated with 4uM DMMB, and illuminated with cool-white fluorescent light. Control and treated samples were then used to infect PHA-stimulated PBMCs and incubated for 7 days, followed by p24 detection by EIA. Similarly, plasma was inoculated with HIV infected PBMC's and treated with 4uM DMMB; centrifuged and washed HIV-infected cells were co-cultured with PHA-stimulated PBMC for 7days followed by p24 measurement.Results: Photo treatment of plasma, packed cells and whole blood inactivated 2000 TCID50 of HIV-1. Also DMBB inactivated intracellular HIV-1, i.e. HIV-infected PBMC in plasma medium was inactivated and cell-to-cell transfer of HIV-1 was prevented.Conclusions: DMMB can inactivate both intra- and extra-cellular HIV-1 under conditions, which minimally alter RBC properties.
