Research Project: ENZYMOLOGY OF POLYKETIDE SYNTHASES FROM SORGHUM BICOLOR

Objective:
The objective of this work is to determine the kinetic parameters for the substrates used by two (2) polyketide synthases that are involved in the biosynthesis of the allelochemical sorgleone.

Approach:
Recombinant enzymes for the two (2) sorghum polyketide synthase genes will be produced by overexpression in E. coli with overexpression vectors developed specifically for this work. These recombinant enzymes will be purified from E. coli cellular lysates using a polyhistidine tag affinity column. Enzyme assays will be performed, and kinetic parameters will be determined by non-linear regression analysis using methods developed by Cooperator.
