Problem: Need an effective system to pre-process samples for nucleic acid analysis (extraction and clean-up).

Project Activity Scope (from Technical Overview Chart): Mechanisms through Instruments

PNNL Solution:

  • Use microbeads with selective surface coatings in a renewable column to capture target analyte
  • Use a scalable automated fluidic system for processing microliters to liters of sample
  • Automatically perfuse sample and wash solutions to remove inhibitory and interfering compounds
  • Elute the concentrated and purified sample suitable for PCR analysis

Capabilities Employed to Meet Client Needs: Optmized fluidics processing protocols, silica and polymer microbeads, reagent optimization, simple automated fluidics system design, sample purification protocols, interferent and inhibitor removal approaches and reagents, polymerase chain reaction (PCR) validation.
