Previously, 8 factor sera were prepared by reciprocal adsorption of polyclonal sera (PS) with whole cells from C. neoformans, serotypes A, B, C, and D. Sera specific for the individual antigenic factors (AF) 5, 6, 7 and 8 have been isolated, but the technics used did not permit for the preparation of sera specific to AF 1, 2, 3, and 4. In this study PS were generated by immunizing rabbits with neoglycoconjugates comprised of GXM from reference isolates from each serotype of C. neoformans and adipic acid dihydrazide derivatives of bovine serum albumin. Immunoaffinity matrices were prepared by the immobilization of GXMs from serotype A, B, C, and D. PS were partially purified using E-Z Sep (Pierce). Anti-serotype C PS, containing Abs to AF 1, 4, and 6 (Factor Serum 1), was applied to a serotype A GXM column. The eluate reacted with the GXMs of all serotypes by dot immunoassay (DEA) and corresponds to AF 1. The effluent from the serotype A GXM column, comprised of AF 4 and 6 (Factor Serum 4), was applied to a serotype B GXM column. The eluate from serotype B GXM column reacted weakly to serotype B GXM (Abs to AF-4), and the effluent reacted strongly to serotype C GXM (Abs to AF-6) by DEA. Similarly, PS against serotype D (AF 1, 2, 3, and 8), serotype A (AF 1, 2, 3, and 7) were fractionated by immunoaffinity chromatography. Abs against AF2 reacted strongly with GXMs from serotypes A, B, and D. Abs against AF3 reacted strongly with serotype D GXM and reacted moderately with serotype A GXM. No activity to AF 7 was detected. Abs against AF 8 reacted strongly and specifically to serotype D GXM. The purified Abs can be used to define epitope structure and for serotyping C. neoformans isolates.
