Immunogenetics of HTLV-II endemic Venezuelan amerindian populations.

Objectives: To define HLA-linked susceptibility to HTLV-II infection among two endemically infected Venezuelan Amerindian tribes, the Guahibo and the Yaruro. Methods: DNA was obtained from the peripheral blood mononuclear cells of 58 Guahibo and 23 Yaruro individuals. SK110/SK111 PCR-oligomer restriction (OR) assay was used for detection of HTLV-II proviral DNA. HLA-DRB1 and DQB1 alleles were defined by a PCR amplification and sequence specific oligonucleotides probes (SSOP) high-resolution method. Results: 15 Guahibo (25%) and 10 Yaruro (43%) were positive for proviral HTLV-IIB sequences. In the Guahibo, DRB1*1402-DQB1*0301 haplotype was highly prevalent, with a higher frequency among HTLV-II carriers (77%) as compared to uninfected individuals (57%). In the Yaruro community this haplotype was also highly prevalent (40%), and additionally, haplotypes DRB1*1406-DQB1*0301 (20%) and DRB1*0403-DQB1*0302 (20%) were also frequent. Conclusions: No significant differences were found in allelic or haplotypic frequencies when infected and non-infected subjects were compared, in both communities. Two of the haplotypes previously associated with HTLV-I infection in South American Indians were highly prevalent in the Yaruro community. Further studies are needed to clarify whether susceptibility to HTLV-I/II infection is in fact related to the HLA system.
