In vitro activity of antifungal agent LY303366 against Candida species, other yeasts and Aspergillus species.

LY303366 (LY) is a new echinocandin B antifungal agent. The activity of LY was compared with amphotericin (AM), 5-fluorocytosine (FC), itraconazole (IT), ketoconazole (KT), and fluconazole (FZ), against 172 clinical strains of 10 Candida spp., 20 strains of 5 non-Candida species and 13 strains of Cryptococcus spp. Strains were tested by broth macro and/or microdilution according to NCCLS document M27-T, with Alamar Blue incorporated into the medium to facilitate the reading of endpoints. RPMI was used for 52 strains of 7 Aspergillus spp. The MIC(50) and MIC(90) (micrograms/ml) for Candida spp. tested were: (Table: see text) C. parapsilosis was the least susceptible to LY. LY was not active against the non-Candida species tested or against Cryptococcus spp. For the Aspergillus spp., two breakpoints for LY were observed; the first gave an MIC(90) for all species of 0.04 micrograms/ml, and the second breakpoint gave an MIC(90) of 5 micrograms/ml. The MIC(90) for AM with Aspergillus spp. was 2 micrograms/ml and for FZ was greater than or equal to 64 micrograms/ml. All species of Aspergillus tested had similar MICs for LY. These results indicate that LY has excellent in vitro activity against most species of Candida associated with clinical infections, and has good to moderate activity against several species of Aspergillus.
