Detection of human papillomavirus DNA in semen by nested PCR.

OBJECTIVE: Human papillomavirus (HPV) infections of the genital tract are sexually transmitted and an association with benign and malignant proliferated lesions in both sexes has been suggested. In order to investigate the possibility of HPV transmission via semen we have tested semen for presence of HPV DNA. METHODS: Semen were obtained by masturbation after voiding from eight patients with condyloma acuminatum on penis, 16 husbands of patients with cervical cancer and 35 infertile males. HPV was detected by nested PCR which was second rounds of PCR amplification with consensus primer pairs for HPV-6, 11, 16, 18, 31, 33, 52 and 58. The HPV types were determined by restriction enzyme digestion. RESULTS: Of six patients with condyloma acuminatum that were positive for HPV 6, four (66.7%) had identical type of the HPV in their semen. Of two patients with condyloma acuminatum that were positive for HPV 11, one (50%) had identical type of the HPV in his semen. HPV was not detected in semen from 16 husbands of patients with cervical cancer and 35 infertile patients. In order to exclude the existence of HPV in the urethra of eight patients with condyloma acuminatum, we tested urine before masturbation and HPV was not detected. DISCUSSIONS AND CONCLUSIONS: The nested PCR technique is a sensitive procedure for the detection of HPV and can detect 10(-2) copy of HPV DNA per cell. HPV was detected in the semen of patients with condyloma acuminatum and shown to be the same type as in the tissue of condyloma acuminatum. The data sugested that the prostate or seminal vesicle might act as a reservoir of HPV and the transmission of HPV might be possible as via semen as by direct contact transmission from infected lesion.
