Labeling, Isolation, and Microarray Analysis of Nascent Versus Steady State RNA via Bromouridine Following Serum Deprivation
 
The response following serum starvation in human foreskin fibroblasts is a well studied classical model of gene expression. After inducing human foreskin fibroblasts to enter a quiescent state through serum starvation for 48 hours, fibroblasts were given normal serum levels. Through addition of bromouridine (BrU) at various time points after reintroducing serum, nascent RNA transcribed within discrete time windows was labeled. Furthermore, immunocytochemical work suggests that such BrU-labeled nascent RNA can be isolated using anti-BrdU. This also suggests that comparisons between nascent and steady state RNA are possible.
