Description/Abstract The Mls-reactive, murine helper T cell clone, designated L2, both proliferates and secretes lymphokines including IL-2 and interleukin-3 (IL-3) when stimulated with alloantigen or with a combination of phorbol myristate acetate (PMA) and the divalent cation ionophore A23187.^L2 cells were exposed to a low concentration (1 Unit(U)/ml) or to a high concentration (50-100 U/ml) of recombinant human IL-2 and cultured for 48-72 hours prior to restimulation with antigen or with PMA + A23187.^Compared to L2 cells exposed to a low concentration of rIL-2, cells exposed to a high concentration of rIL-2 were profoundly less responsive to antigen as indicated a 6-16 fold reduction in antigen-stimulated lymphokine secretion.^Consistent with the reduction in antigen-stimulated lymphokine secretion, levels of antigen-stimulated mRNA for both IL-2 and IL-3 were diminished 8-16 fold in L2 cells previously exposed to a high concentration of rIL-2.^In contrast, L2 cells exposed to a high or low concentration of rIL-2 produced similar amounts of IL-2 and IL-3 when stimulated with PMA + A23187.^These data suggest that exposure of L2 cells to a high concentration of IL-2 may inhibit the intracellular pathway which links antigen recognition to the production of lymphokine mRNA.
